Drug class description :  4-quinolones (quinolones).
Generic Name :  Ofloxacin
Drug description :  Tarivid: 200 mg off-white f-c oblong scored tablet marked MXI and logo; 400 mg yellow f-c oblong scored tablet. Tarivid Infusion: solution.
Presentation :  Tarivid: Ofloxacin 200mg , 400mg . Tarivid Infusion: Ofloxacin (as hydrochloride) 2 mg /mL.
Indications :  Tarivid: upper and lower urinary tract infections, lower respiratory tract infections, uncomplicated urethral and cervical gonorrhoea, non- gonococcal urethritis and cervicitis, skin and soft tissue infections. Tarivid Infusion: acute and chronic infections of the lower respiratory tract, acute and chronic upper and lower urinary tract infections, skin and soft tissue infections, septicaemia. Microbiological results indicate that the following organisms may be regarded as sensitive: Staphylococcus aureus (including methicillin resistant staphylococci), Staphylococcus epidermidis, Neisseria species, Escherichia coli, Citrobacter, Klebsiella, Enterobacter, Hafnia, Proteus (indole-negative and indole-positive), Salmonella, Shigella, Acinetobacter, Yersinia enterocolitica, Campylobacter jejuni, Aeromonas, Plesiomonas, Vibrio cholerae, Vibrio parahaemolyticus, Haemophilus influenzae, Chlamydiae, Legionella, Gardnerella.Variable Streptococci, Serrratia marcescens, Pseudomonas aeruginosa, Clostridium species and Mycoplasmas.Anaerobic bacteria are usually resistant. Tarivid is not normally active against Treponema pallidum.
Adult Dosage :  Tarivid: Depending on type and severity of infection, 200 - 800 mg once daily in the morning or larger doses in two divided doses. Lower UTI, 200 - 400 mg daily. Upper UTI, 200 - 400 mg daily, increasing to 400 mg twice daily if necessary. LRTI, 400 mg daily, increasing to 400 mg twice daily if necessary. Gonorrhoea, 400 mg as a single dose. NGU, 400 mg daily. Skin and soft tissue infections, 400 mg twice daily. Tarivid Infusion: Usually 200 - 800 mg daily by slow intravenous infusion over 30 minutes for 7 - 10 days depending on type and severity of infection. Complicated UTI, 200 mg daily. LRTI and septicaemia, 200 mg twice daily. Skin and soft tissue infections, 400 mg twice daily.
Child Dosage :  Not recommended.
Contra Indications :  History of epilepsy or tendinitis. Pregnancy, lactation. Growing adolescents.
Special Precautions :  Psychiatric disorders. Renal and hepatic impairment. Haemodialysis or peritoneal dialysis. 6-G-PD deficiency. Exposure to strong sunlight or UV rays. Driving or operating machinery.
Interactions :  Magnesium or aluminium antacids, sucralfate, glibenclamide, iron, probenecid, cimetidine, frusemide (furosemide), methotrexate, NSAIDs, anticoagulants. Injection: barbiturate anaesthetics, hypotensives.
Adverse Reactions :  GI upset, hypersensitivity reactions, skin reactions, convulsions, CNS disturbances, (discontinue if suspected). Transient increases in liver enzymes, disturbance of kidney function and blood sugars, porphyria, tachycardia, hypotension. Rarely, weakness, joint and muscle pain, tendonitis (discontinue treatment and rest limb), tendon rupture, bone marrow depression, acute interstitial nephritis, serum creatinine increase, cholestatic jaundice, hepatitis, severe liver damage, haematological disturbances. Injection: pain at injection site, thrombophlebitis.
