Drug class description :  GABA analogues (anticonvulsants / anti-epileptics).
Generic Name :  Vigabatrin
Drug description :  Sabril Tablets: white oval scored tablet with tablet name. Sabril Sachet: powder.
Presentation :  Sabril Tablets: Vigabatrin 500 mg . Sabril Sachet: Vigabatrin 500 mg .
Indications :  Adjunctive treatment of partial Adjunctive treatment of partial seizures with or without secondary generalisation in patients inadequately controlled by, or intolerant to, all other anticonvulsant combinations. Monotherapy for infantile spasms.
Adult Dosage :  Initially 1 g daily in single or two divided doses added to existing therapy. Adjust dose in 500 mg weekly increments according to response; maximum 3 g daily.
Child Dosage :  Initially 40 mg/kg daily. Maintenance, see data sheet. Infantile spasms, initially 50mg/ kg daily titrated over a one week period if necessary. Doses up to 150 mg/kg daily may be required.
Contra Indications :  Pre-existing visual field defect. Lactation.
Special Precautions :  History of psychosis or behavioural problems. Elderly, renal impairment. Monitor neurological function. Test visual field before and during therapy; advise patient to report new visual problems - if symptoms develop refer to ophthalmologist and consider discontinuation. Withdraw gradually over 2 - 4 weeks. Pregnancy.
Interactions :  Phenytoin. ALT and AST liver tests.
Adverse Reactions :  Visual field defects. Agitation, aggression, nervousness, irritability, depression, memory and thought disturbances, paranoid reaction, psychosis, somnolence, fatigue, headache, weight gain, tremor, oedema, dizziness, paraesthesia, sedation, confusion with EEG changes, GI upset, blurred vision, diplopia, nystagmus. Excitation and agitation in children. Increased frequency of seizures in patients with myoclonic seizures.
