INJEKSJONSVÆSKE 2 mg/ml: 1 ml inneh.: Doxorubicin. hydrochlorid. 2 mg, acid. hydrochloric. q.s. ad pH 3, natr. chlorid. 9 mg et aqua ad iniect. ad 1 ml. PULVER TIL INJEKSJONSVÆSKE 10 mg og 50 mg: Hvert hetteglass inneh.: Doxorubicin. hydrochlorid. 10 mg, resp. 50 mg, lactos. 50 mg, resp. 250 mg, methyl. parahydroxybenz. (E 218) 1 mg, resp. 5 mg. |
| Indications : | Brystcancer. Osteogent sarkom og Ewings sarkom. Bløtdelssarkom. Småcellet anaplastisk lungekarsinom. Malignt lymfom både av typen mb. Hodgkin og non-Hodgkin. Urogenitale og gynekologiske tumores som blærecancer, testiscancer og uteruscancer. Pediatriske solide tumores som rhabdomyosarkom, neuroblastom, Wilms tumor. Akutte leukemier. Forsøksvis ved: Prostatakarsinom. Thyreoideakarsinom. Ovariecancer, cervixcancer, vaginalcancer, «head and neck»-cancer, ventrikkelcancer. |
| Adult Dosage : | Skal alltid gis gjennom venekanyle til en frittløpende væskeinfusjon (fysiologisk saltvann) i løpet av 2-5 minutter 60-75 mg/m 2 kroppsflate som engangsdose i.v. med 21 dagers intervall. Den laveste dosen gis til pasienter med nedsatt benmargsfunksjon. Alternativ dosering: 30 mg/m 2 kroppsflate/dag i 3 dager med 4 ukers mellomrom, eller ved cancer mammae: 20 mg pr. uke i.v. over 3-4 minutter. For å unngå kardiomyopati skal totaldosen ikke overstige 550 mg/m 2 . Innen behandlingen innledes, anbefales en kontroll av leverfunksjonen ved hjelp av konvensjonelle tester som ASAT, ALAT, ALP og bilirubin. Ved nedsatt leverfunksjon skal dosen reduseres etter følgende tabell: Gitt i kombinasjon med andre cytostatika er normaldosen 30-60 mg/m 2 kroppsflate. Intravesikal instillasjon har vært forsøkt i forbindelse med blærecancer, se spesiallitteratur. |
| Child Dosage : | Cytostatikum. |
| Elderly Dosage : | Overgang i placenta: Bør ikke gis under graviditet. Spesielt i 1. trimester må morens behov nøye veies mot risikoen for fosterskade. |
| Contra Indications : | Skal ikke gis til pasienter med uttalt myelosuppresjon indusert av tidligere behandling med andre cytostatika eller ved strålebehandling. Graviditet: Se Graviditet/Amming. |
| Special Precautions : | Forsiktighet bør utvises hos pasienter med hjertesykdom i anamnesen, og hos pasienter som har fått strålebehandling mot mediastinum. Det samme gjelder hvis andre kardiotoksiske substanser som f.eks. daunorubicin er gitt tidligere eller potente hjertetoksiske preparater gis samtidig med doksorubicin. |
| Adverse Reactions : | Svært høye doser kan forårsake myelosuppresjon. Kombinasjonsbehandling skal da iverksettes for å hjelpe, og omfatter da ev. antibiotikabehandling, blodtransfusjoner og kullfilterdialyse. |
